摘要: Single nucleotide polymorphisms (SNPs) which represent the most widespread source of sequence variation in genomes, are becoming a routine application several fields such as forensics, ecology and conservation genetics. Their use, requiring short amplifications, may allow more efficient genotyping degraded DNA. We provide first applica- tion SNP an Italian non-invasive genetic monitoring project wolf. compared three different techniques for SNPs: pyrosequencing, SNaPshot ® TaqMan Probe Assay real-Time PCr . succes- sively genotyped nine SNPs using 51 wolves, 57 domestic dogs, 15 wolf x dog hybrids 313 scats collected northern Apennines. The obtained results were used to estimate variability error rates protocols standard microsatellite analysis. evaluated cost, labora- tory effort reliability these markers discuss possible future use VeraCode, SNPlex Fluidigm EP1 system wild population monitoring.
