摘要: For Mycobacterium tuberculosis, phenotypic methods for drug susceptibility testing of second-line drugs are poorly standardized and technically challenging. The Sensititre MYCOTB MIC plate (MYCOTB) is a microtiter containing lyophilized antibiotics configured determination MICs to first- antituberculosis drugs. To evaluate the performance M. tuberculosis using Middlebrook 7H10 agar proportion method (APM) as comparator, we conducted two-site study archived isolates from Uganda Republic Korea. Thawed were subcultured, dilutions inoculated into wells onto agar. results read at days 7, 10, 14, 21; APM 21 days. A total 222 provided on both platforms. By APM, 106/222 (47.7%) resistant least isoniazid rifampin. Agreement between with respect or resistance was ≥92% 7 12 when strict definition used ≥96% 10 agreement defined by allowing ± one-well range around critical concentration. ethambutol, 80% 81%. moxifloxacin, 83% 85%; incorporating existing DNA sequencing information discrepant analysis raised 91% 96%. MYCOTB, median time interpretation interreader ≥95% all reliable except available sooner than those determined APM.
