摘要: Abundant evidence now supports the existence of phospholipids in nucleus that resist washing nuclei with detergents. These lipids are apparently not nuclear envelope as part a bilayer membrane, but actually within form proteolipid complexes unidentified proteins. This review discusses experimental attempts to explain their existence. Among these polyphosphoinositol which, together enzymes synthesize them, an intranuclear phospholipase C (PI-PLC) signaling system generates diacylglycerol (DAG) and inositol 1,4,5-trisphosphate [Ins(1,4,5)P3]. The isoforms PI-PLC involved this system, how they regulated, yet entirely clear. Generation DAG is believed recruit protein kinase (PKC) phosphorylate Ins(1,4,5)P3 may mobilize Ca2+ from space between membranes thus increase nucleoplasmic Ca2+. Less well understood increasing number variations complications on "simple" idea system. include, all nucleus, (i) two routes synthesis phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2]; (ii) sources DAG, one pathway other probably phosphatidylcholine; (iii) several PKC translocating nuclei; (iv) increases activity at points cell cycle; (v) phosphorylation Ins(1,4,5)P3, which have functions, including role transfer mRNA out nucleus; (vi) possible lipid pathways include sphingolipids, A2, and, particular, 3-phosphorylated lipids, emerging major players signaling.
