Myogenic progenitors and imaging single-cell flow analysis: a model to study commitment of adult muscle stem cells

摘要: Research on skeletal muscles suffers from a lack of appropriate human models to study muscle formation and regeneration the regulatory level single cells. This hampers both basic understanding development new therapeutic approaches. The use imaging multicolour flow cytometry myogenic stem cells can help fill this void by allowing researchers visualize quantify reaction individual cultured bioactives or other physiological impulses. As proof concept, we subjected CD56+ satellite reference follistatin Malva sylvestris extracts then used multicolor stepwise activation factors MyoD myogenin in approach enabled us evaluate potency these stimulate commitment. To validate method, multi-photon confocal microscopy confirm potential differentiation expression desmin. Imaging revealed statistically significant differences between treated untreated groups progenitors propose utilization concept as an integral part future research strategies.
