摘要: IS629 is 1,310 bp in length with a pair of 25-bp imperfect inverted repeats at its termini. Two partially overlapping open reading frames, orfA and orfB, are present IS629, two putative translational frameshift signals, TTTTG (T4G) AAAAT (A4T), located near the 3′-end orfA. With lacZ gene as reporter, both T4G A4T motifs determined to be −1 signal. peptides representing transframe products designated OrfAB′ OrfAB, identified by liquid chromatography-tandem mass spectrometric approach. Results transposition assays show that transposase OrfAB aids IS629. Pulse-chase experiments Escherichia coli two-hybrid demonstrate binds stabilizes OrfAB′, thus increasing activity This first transposable element IS3 family shown have functional frameshifted involved use product regulate transposition.
