摘要: : Rats were administered [3H]fucose by intracranial injection and synaptic membranes (SMs) isolated 18 h later. Oligosaccharides associated with SM glycoproteins prepared hydrazinolysis analyzed a combination of affinity chromatography on concanavalin A (Con A)-agarose, ion-exchange DEAE-celluiose, gel permeation chromatography. Most (94%) the [3H]fucose-labelled oligosaccharides present in fraction that did not bind to Con A. Of these 41% DEAE-cellulose, indicating absence negatively charged groups remainder resolved into four fractions increasing acidity. Gel from DEAE-cellulose column suggested major corresponded fucosylated triantennary structures containing varying amounts sialic acid although more highly branched peripheral branches lacking one or sugars may also have been present. Comparison fucosyl SMs 10- 28-day-old animals indicated general oligosaccharide content was similar at both ages, younger characterized an increase proportion acidic structures. Fucosylated glycans derived junctional (SJ) greater percentage than SMs. The results indicate SJs are specific complements glycoprotein oligosaccharides.
