摘要: Abstract The mechanisms for endothelial cell injury induced by the lipid hydroperoxide 15-hydroperoxyeicosatetraenoic acid (15-HPETE), an arachidonate lipoxygenase product, were explored in cultured bovine cells. In serum-free medium, there was significant incorporation of [3H]-15-HPETE into phospholipids monolayers, and 15-HPETE severe injury, which determined 51Cr-release assay. contrast, serum containing little cells, no cellular occurred. free condition, mainly incorporated phospholipids. produced peroxidation, accumulation malondialdehyde 15-HPETE-induced peroxidation suppressed radical scavengers (MK-447, MCI-186), anti-oxidants (α-tocopherol, butylated hydroxytoluene) iron chelators (desferrioxamine,22′-bipyridine). Furthermore, these agents also cytotoxicity. These results indicate that depends on iron-mediated peroxidation.
