摘要: Methods for processing samples which may contain mycobacteria are compatible with both conventional culturing techniques and nucleic acid analysis. It has been found that the harsh chemical treatment previously thought necessary to obtain efficient lysis can be eliminated without loss of efficiency, thereby eliminating reagents inhibit subsequent acid-based reactions. Heat alone is sufficient lyse mycobacteria, even in buffers do not detergents, chelators, enzymes, etc. Residual introduced by sample methods culture successfully removed from processed pellet significant microorganisms at least two washings saline, water, or analysis appropriate centrifugation.
