Immunomodulation of phloretin by impairing dendritic cell activation and function.

摘要: Dietary compounds in fruits and vegetables have been shown to exert many biological activities. In addition antioxidant effects, a number of flavonoids are able modulate inflammatory responses. Here, we demonstrated that phloretin (PT), natural dihydrochalcone found fruits, suppressed the activation function mouse dendritic cells (DCs). Phloretin disturbed multiple intracellular signaling pathways DCs induced by Toll-like receptor 4 (TLR4) agonist lipopolysaccharide (LPS), including ROS, MAPKs (ERK, JNK, p38 MAPK), NF-κB, thereby reducing production cytokines chemokines. also effectively treated with different dosages LPS or various TLR agonists. The LPS-induced DC maturation was attenuated because expression levels MHC class II co-stimulatory molecules were down-regulated, which then inhibited LPS-stimulating subsequent naive T cell mixed lymphocyte reaction. Moreover, vivo administration phenotypic LPS-challenged splenic decreased IFN-γ from activated CD4 cells. Thus, suggest may potentially be an immunomodulator impairing phloretin-contained helpful improvement inflammation autoimmune diseases.
