摘要: There is an urgent need for in vitro screening assays to evaluate nanoparticle (NP) toxicity. However, the relevance of still disputable. We administered doses TiO(2) NPs different sizes alveolar epithelial cells and same by intratracheal instillation rats vivo examine correlation between responses. The correlations were based on toxicity rankings after adopting NP surface area as dose metric, response per unit metric. Sizes anatase ranged from 3 100 nm. A cell-free assay measuring reactive oxygen species (ROS) was used, lactate dehydrogenase (LDH) release, protein oxidation induction cellular using a rat lung Type I cell line (R3/1) following 24 h incubation. endpoint number PMNs bronchoalveolar lavage fluid (BALF) exposure via instillation. Slope analyses curves shows that responses well correlated. conclude approach steepest slope analysis offers superior method correlate with results ranking their toxic potency.
