Asymmetric Segregation of the Tumor Suppressor Brat Regulates Self-Renewal in Drosophila Neural Stem Cells

摘要: How stem cells generate both differentiating and self-renewing daughter is unclear. Here, we show that Drosophila larval neuroblasts-stem cell-like precursors of the adult brain-regulate proliferation by segregating growth inhibitor Brat transcription factor Prospero into only one cell. Like Prospero, binds cosegregates with adaptor protein Miranda. In neuroblasts, are required to inhibit self-renewal in two cells. While regulates cell-cycle gene transcription, acts as a posttranscriptional dMyc. brat or prospero mutants, grow behave like neuroblasts leading formation brain tumors. Similar defects seen lethal giant larvae (lgl) mutants where not asymmetric. We have identified molecular mechanism may control prevent tumor other well.
