摘要: To identify structures that determine the 90° orientation of thin espalier dendritic trees Purkinje cells with respect to parallel fibers (axonal neurite bundles granule cells) in cerebellar cortex, we designed five types two-dimensional and three-dimensional cell tissue cultures cerebella from postnatal mice analyzed dendrites astrocyte by immunofluorescence double or triple staining. We cultured dissociated on micropatterned substrates preformed a microexplant culture two-dimensionally matrix gels three-dimensionally. Dendrites, but not axons, extended toward neurites oriented at right angles aligned three cultures. In more organized explant proper culture, only consistently also two-dimensionally. However, “organotypic culture,” which three-dimensionally thick mimicking were produced, often perpendicular Astrocytes abundant all cultures, there was no definite correlation between presence dendrites, although their frequently associated organotypic culture. Therefore, may grow newly produced cortex be mainly via “perpendicular contact guidance.” J. Comp. Neurol. 499:274–289, 2006. © 2006 Wiley-Liss, Inc.
