Effects of Irradiation on the Release of Growth Factors from Cultured Bovine, Porcine, and Human Endothelial Cells

摘要: The effects of radiation on the release mitogenic factors into media cultured endothelial cells bovine, porcine, and human origins were studied. Although unirradiated controls revealed a significant background activity, single doses irradiation (20–60 Gy) resulted in dose-related increased growth factor measured by conditioned both 3T3 mouse fibroblasts serving as target cells. Receptor binding competition assays for platelet-derived receptor that 12–28% total activity was due to factor-like mitogens. Mitogenic using specific antibody mediated inhibition suggested another component fibroblast factor. increase cell death, enhanced activities did not appear result from lysis-related leakage intracellular stores factors. Instead, our data suggest synthesized de novo secreted at elevated levels which maintained postradiation high level metabolic activity. Time course studies demonstrated accumulation started within first 24 h after reached plateau 72 treatment.
