摘要: Clathrin-coated vesicles mediate endocytosis and transport between the trans-Golgi network (TGN) endosomes in eukaryotic cells. Clathrin adaptors play central roles coat assembly, interacting with clathrin, cargo membranes. Two main types of clathrin adaptor act TGN-endosome traffic: GGA proteins AP-1 complex. Here we characterize relationship proteins, other TGN using live-cell super-resolution microscopy yeast. We present evidence that are recruited sequentially two waves assembly at TGN. Mutations decrease phosphatidylinositol 4-phosphate (PtdIns(4)P) levels slow or uncouple from assembly. Conversely, enhanced PtdIns(4)P synthesis shortens time waves. Gga2p binds directly to PtdIns(4)-kinase Pik1p contributes recruitment. These results identify a PtdIns(4)P-based mechanism for regulating progressive adaptor-specific coats
