摘要: Scanning force microscopy was used for mapping the viscoelastic properties of metaphase chromosomes. These were probed by scanning with various imaging forces and subsequent calculation difference image. The procedure allows a behavior expressed as force-dependent indentation local surface feature results in an image material contrast. approach is demonstrated on rehydrated chromosomes, which spread air-dried before rehydration aqueous buffer. resulted swelling chromosome structure accompanied drastic changes properties.
