Molecular cloning of a candidate tumor suppressor gene, DLC1, from chromosome 3p21.3.

摘要: The short arm of chromosome 3 is thought to contain multiple tumor suppressor genes, because one copy this chromosomal frequently missing in carcinomas that have arisen a variety tissues. We isolated novel gene encoding 1755-amino acid polypeptide, through large-scale sequencing genomic DNA at 3p21.3. Mutational analysis by reverse transcription-PCR revealed the lack functional transcripts and an increase nonfunctional RNA significant proportion (33%) cancer cell lines primary cancers (4 14 esophageal lines, 2 renal 11 30 non-small lung cancers, 10 squamous esophagus). However, no alterations itself were detected any examined. Introduction cDNA significantly suppressed growth four different two which produced normal transcript on their own. No such effect occurred when antisense cDNA, corresponding aberrant transcript, or vector alone transfected. These data suggest transcription gene, designated DLC1 (deleted 1), may be involved carcinogenesis lung, esophagus, kidney.
