Anti-cancer effect of rubropunctatin against human gastric carcinoma cells BGC-823

摘要: The Monascus pigment, rubropunctatin, was extracted and purified from red mold rice (RMR) its cytotoxic activities against human gastric adenocarcinoma BGC-823 cells were studied both in vitro vivo. Rubropunctatin inhibited the proliferation of with an inhibitory concentration (IC50) 12.57 μM, while it exhibited no significant toxicity to normal epithelial cell GES-1 at same concentration. Treatment rubropunctatin resulted a dose- time-dependent apoptosis, as validated by increase percentage sub-G1 phase phosphotidylserine externalization. vivo experimental data demonstrated that could offer similar therapeutic benefits comparison dose taxol. After five times intravenous injection, tumor weight BGC-823-bearing nude mice reduced 23.5% 8 mg/kg 37.7% 32 mg/kg, respectively. expressions 30 genes related induction apoptosis found up-regulated significantly. two most expressed necrosis factor (TNF) DNA-damage inducible transcript 3. TNF considered major mediator induced rubropunctatin. This is first report describing anti-proliferative effect mechanism on cells. has potential be developed new natural anti-cancer agent.
