摘要: Abstract In cultures of normal adult human glia-like cells, density-dependent cell cycle inhibition (topoinhibition) and contact ruffling occur almost simultaneously, suggesting a functional coupling between activities the surface initiation DNA synthesis. The present paper examines whether cytochalasin B (CB), which reversibly inhibits ruffling, also blocks glia cycle. effects drug (2 μg/ml) were following: 1. Initiation synthesis subcultivated stationary cells was inhibited. 2. Stimulation in by medium change suppressed. 3. Migration into wound confluent layer blocked as well lining wound. 4. (but not continuation) exponentially growing inhibited leading to population mainly arrested G 1 determined microspectrophotometry on Feulgen-stained cells. Topoinhibited Since cytokinesis CB, fraction binuclear appeared. block induced CB reversible, even after several weeks treatment, with exception that more reluctantly entered S phase release block. conclusion, efficiently induces reversible probably physiologic This finding strengthens notion connection membrane proliferation. underlying mechanism is discussed.
