摘要: Insulin-like growth factors (IGFs) are often essential for the maintenance of malignant phenotype, and in lung cancer IGF-I receptor (IGF-Ir) is expressed at high levels. Stable transfection antisense plasmids expressing first 300 bp IGF-Ir reduces tumorigenicity a variety tumor cell lines has been reported to induce systemic antitumor effects on established, non-gene-modified tumors animal model systems. We have constructed an adenovirus (Ad-IGF-Ir/as) attempt develop these observations into clinical therapeutic approach. A single transduction by Ad-IGF-Ir/as (at multiplicity infection 10:1) decreased number about 50% human NCI H460 SCC5, as measured 125I-labeled competitive binding assay. After Ad-IGF-Ir/as, soft agar clonogenicity was reduced 84%. The i.p. treatment nude mice bearing established cells resulted prolonged survival compared that treated with reporter virus. These results suggest effect xenografts may represent effective practical gene therapy strategy.
