Generation of Viable Embryos and Embryonic Stem Cell-Like Cells from Cultured Primary Follicles in Mice

摘要: Primary follicles retrieved from B6CBAF1 prepubertal mice were cultured in a stepwise manner an alpha-minimum essential medium-based medium to generate viable embryos and embryonic stem cell (ESC)-like cells. A significant increase follicle growth oocyte maturation accompanied by increased secretion of 17beta-estradiol progesterone was achieved exposing primary 100 or 200 mIU follicle-stimulating hormone (FSH) during culture. More oocytes developed into blastocysts following vitro fertilization (IVF) parthenogenetic activation after culture with FSH the entire period than mIU. Eleven ESC-like lines, consisting four heterozygotic seven homozygotic phenotypes, established 25 trials combined IVF activation. In conclusion, can potentially yield developmentally competent oocytes, which produce lines manipulation. We suggest method utilize immature follicles, are most abundant ovaries, improve reproductive efficiency for use regenerative medicine.
