Melatonin and Metformin Diminish Oxidative Stress in Heart Tissue in a Rat Model of High Fat Diet and Mammary Carcinogenesis.

摘要: The aim of this study was to determine the effects long-term administration oral antidiabetic metformin or pineal hormone melatonin, and a combination thereof, in preventing oxidative stress heart tissue female Sprague-Dawley rats with mammary tumors induced by N-methyl-N-nitrosourea (NMU) (50 mg/kg) given on 42nd postnatal day. Metformin melatonin were administered 12 days before 16 weeks after carcinogen. During experiment, all animals fed high fat diet (10% total fat, 2.5% from lard, 7.5% palm oil). findings are that carcinogenesis generated stress. Reactive oxygen species (ROS) content, estimated thiobarbituric acid reactive substances (TBARS), oxidatively modified protein content (aldehyde ketone derivatives), activity antioxidant enzymes superoxide dismutase, glutathione reductase, peroxidase augmented. caused decrease heart, accompanied diene conjugates, elimination ROS (stable status), activation catalase reductase. Melatonin an increase status substantial reduction as aldehyde derivatives, lipid peroxidation at initial (diene conjugates) terminal stages increased activities. combined reversed NMU In conclusion, reduces level tissue, diet, significantly stronger than metformin.
