Detection of Australian Field Isolates of Citrus Tristeza Virus by Double‐Stranded RNA Analysis

摘要: Double-stranded RNA analysis indicated that widespread latent infection by citrus tristeza virus (CTV) occurs naturally in a variety collection at Merbein, Australia. Detection was based on the presence of putative replicative form (RF) (ca 19. 5 kilobase pairs) and or absence three previously known subgenomic dsRNAs with molecular sizes ca 3. 1,1.7 0.8 pairs. With exception some relatives normally used as rootstocks, were readily detectable all trees tested. Variability noticed among profiles individual seedling Microcitrus australasica var. sanguinea. By contrast, dsRNA patterns open-pollinated Ellendale tangor indistinguishable. A number bands found West Indian lime infected CTV not detected when this isolate graft-inoculated to healthy sweet orange seedlings. simple method for enhancing sensitivity detection silver staining is described which involves incubating gels RNase prior staining; allowed RF only 40 mg green hark material.
