摘要: Genome analysis of over 70 Saccharomyces strains revealed the existence five groups genetically diverged S. cerevisiae wild-type isolates, which feature distinct genetic backgrounds and reflect natural diversity existing among species. The originated from different geographical ecological niches (Malaysian, West African, North American, Wine/European Sake) represent clean, non-mosaic lineages cerevisiae, meaning that their genomes differ essentially by monomorphic private SNPs. In this study, one representative strain for each clean was selected mutated several auxotroph genes markerless deletions, so all dominant markers remained available further manipulations. A set 50 assembled, including eight haploid two diploid lineage. These carry combinations leu2∆0, lys2∆0, met15∆0, ura3∆0 and/or ura3∆::KanMX-barcoded deletions with marker configurations resembling BY series, will allow large-scale crossing deletion collections. This new tractable provides a powerful tool kit to explore impact variation on complex biological processes.
