Effects of prunetin on the proteolytic activity, secretion and gene expression of MMP-3 in vitro and production of MMP-3 in vivo.

摘要: We investigated whether prunetin affects the proteolytic activity, secretion, and gene expression of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well vivo production MMP-3 rat knee joint to evaluate potential chondroprotective eff ect prunetin. Rabbit chondrocytes were a monolayer, reverse transcription-polymerase chain reaction (RT-PCR) was used measure interleukin-1β (IL-1β)-induced MMP-3, MMP-1, MMP-13, disintegrin metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5. In effects on IL-1β-induced secretion activity using western blot analysis casein zymography, respectively. The protein also examined vivo. results follows: (1) inhibited ADAMTS-4, ADAMTS-5; (2) MMP-3; (3) suppressed These suggest that can regulate expression, by directly acting chondrocytes.
