摘要: Abstract Ruthenium(III) complexes are increasingly attracting the interest of researchers due to their promising pharmacological properties. In present study, we investigated ability cis-(dichloro)tetrammineruthenium(III) chloride produce lethal effects in human chronic myelogenous leukemia K562 cells. The MTT tetrazolium reduction test and trypan blue exclusion assay revealed that IC50 for compound after 48 h incubation with cells was approximately 10.74 73.45 μM, respectively. Interestingly, it observed this exhibits mild cytotoxicity towards MRC-5 fibroblast (IC50 > 383 μM). Flow cytometric analysis capable change cell cycle distribution since percentage G1, S G2 phases decreased. addition, treatment induced apoptotic death cells, demonstrated by increased DNA content sub-G1-peak a significant increase caspase-3 activity. Assay using cyclosporin A, an inhibitor mitochondrial permeability transition pore (MPT) showed preincubation had not effect on activation. summary, displayed cytotoxic through arrest induction which suggests might have therapeutic potential against leukemia.
