Allelotyping analyses of synchronous primary and metastasis CIN colon cancers identified different subtypes

摘要: In colorectal cancer, the molecular alterations that lead to metastasis are not clearly established, probably because of their high genetic complexity. To identify combinations changes involved in tumor progression and metastasis, we focused on chromosome instable (CIN) colon cancers. We compared by allelotyping 33 microsatellites, genomic 38 primary tumors with synchronously resected matched liver metastases (CLM). observed (i) number patients at certain loci did differ significantly between whole paired CLM, (ii) a group had fewer when tumor. A 2-way hierarchical unsupervised clustering data revealed 2 subtypes have different levels CIN (CIN-High, CIN-Low). Both minimal common set chromosomes 8p, 17p 18q, but does include alteration 5q or mutation K-Ras. These were also using collection 104 independent tumors. addition, found third subtype, consisting very low associated specific (CIN-Very Low). carcinogenesis may require that, contrast current hypothesis, level correlate progression. Therefore, our results suggest potential could be present early stages development.
