Metastatic potential of human colorectal carcinoma SW1222 cells transfected with cDNA encoding carcinoembryonic antigen

摘要: In order to examine a role of carcinoembryonic antigen (CEA) in metastasis, cDNA encoding CEA was introduced into clone human colorectal carcinoma SW1222 cells. Western blot analysis revealed that all transfectants express 180 kDa while the parent does not. transfectants, level expression 3 higher than 1. Clone formed aggregates rapidly after suspended by trypsinization 1 did experimental metastasis assay where tumor cells were injected intrasplenically, exhibited liver-metastatic activity Fab fragment anti-CEA antibody significantly inhibited both cell aggregation and liver metastases caused 3. These findings suggested expressed on surface may play an important hepatic from carcinoma, possibly through its adhesion activity.
