摘要: Expression of chalcone synthase (CHS), the first enzyme in flavonoid branch phenylpropanoid biosynthetic pathway plants, is induced by developmental cues and environmental stimuli. We used plant transformation technology to delineate functional structure French bean CHS15 gene promoter during development. In absence an efficient procedure for bean, Nicotiana tabacum was as model plant. activity, evaluated measurements β-d-glucuronidase (GUS) revealed a tissue-specific pattern expression similar that reported CHS genes bean. GUS activity observed flowers root tips. Floral confined pigmented part petals transient fashion. Fine mapping cis-elements accomplished using set mutants generated unidirectional deletions or site-directed mutagenesis. Maximal floral root-specific found require sequence elements located on both sides TATA-box. Two adjacent motifs, G-box (CACGTG) H-box (CCTACC(N)7CT) near TATA-box, were essential expression, also be important expression. The regulated complex interplay between different their cognate factors. conservation promoters emphasizes importance regulatory motifs.
