Transcriptional regulation of amelotin gene by proinflammatory cytokines in gingival fibroblasts

摘要: Amelotin (AMTN) is a secreted protein expressed during the late stages of enamel formation and in junctional epithelium. Among many differentially genes, we found significantly increased AMTN mRNA level inflamed gingiva by DNA microarray. The inductions expressions human gingival fibroblasts (HGF) were confirmed real-time polymerase chain reaction. To determine molecular basis expression its regulation proinflammatory cytokines, have isolated characterized promoter region mouse gene. Transient transfection assays performed using luciferase constructs including gene promoter. Interleukin-1β, Interleukin-6 tumor necrosis factor-α induced levels HGF. These cytokines activities results suggest that induce transcription role for inflammation.
