Mural granulosa cell gene expression associated with oocyte developmental competence

摘要: Background: Ovarian follicle development is a complex process. Paracrine interactions between somatic and germ cells are critical for normal follicular oocyte maturation. Studies have suggested that the health function of granulosa cumulus may be reflective status enclosed oocyte. The objective present study to assess, using an in vivo immature rat model, gene expression profile cells, which linked developmental competence We hypothesized specific genes correlated with Methods: Immature rats were injected eCG 24 h thereafter anti-eCG antibody induce atresia or pre-immune serum stimulate development. A high percentage (30-50%, competence, NDC) oocytes from eCG/pre-immune group developed term after embryo transfer compared those eCG/anti-eCG (0%, poor PDC). Gene profiles mural above oocyte-collected follicles assessed by Affymetrix whole genome array. Results: result showed twelve up-regulated, while one was down-regulated more than 1.5 folds NDC PDC group. ontology classification up-regulated included lysyl oxidase (Lox) nerve growth factor receptor associated protein 1 (Ngfrap1), important regulation protein-lysine 6-oxidase activity, apoptosis induction, respectively. glycoprotein-4-beta galactosyltransferase 2 (Ggbt2), involved extracellular matrix organization biogenesis. Conclusions: data demonstrate close association oocytes. This finding suggests most differentially expressed gene, oxidase, candidate biomarker useful selection good quality assisted reproduction.
