Persistent RNA virus infection of lepidopteran cell lines: Interactions with the RNAi machinery

摘要: RNAi is broadly used as a technique for specific gene silencing in insects but few studies have investigated the factors that can affect its efficiency. Viral infections potential to interfere with through their production of viral suppressors (VSRs) and small RNAs saturate inactivate machinery. In this study, impact persistent infection RNA viruses Flock house virus (FHV) Macula-like (MLV) on efficiency was selected lepidopteran cell lines. Lepidopteran lines were found be readily infected by both without any apparent pathogenic effects, exception Bombyx-derived Bm5 BmN4 cells, which could not FHV. Because Sf21 cells free from FHV MLV Hi5-SF only contained low levels MLV, they tested evaluate presence virus. Two types reporter assays however did detect significant interference when compared virus-free cells. Hi5 easily cleared expression an hairpin targets VSR gene, confirming mechanism inhibited. Sequencing indicated B2 inhibitor putative intact persistently lines, indicating protection against remains essential survival. It proposed are too encoded VSRs act locally at sites replication (mitochondrial membranes) affecting rest cytoplasm.
