摘要: In trypanosomatids, transcription is polycistronic and gene expression control occurs mainly at the post-transcriptional level. To investigate role of sequences present in 3′UTR stage-specific mRNAs Trypanosoma cruzi, we generated a new vector, named pTcDUALuc, containing firefly Renilla luciferase reporter genes. test this derived from plus intergenic regions alpha tubulin gene, which up-regulated epimastigotes, amastin, amastigotes, were inserted downstream activity was compared transient stable transfected parasites. As expected, increased detected epimastigotes transiently with pTcDUALuc sequences. Using cell lines that allowed to differentiate into observed mRNA levels amastigotes amastin We also showed spliced leader sequence poly-A tail predicted sites deletions resulted decreased because it affects polyadenylation. contrast constructs genes, presence trans-sialidase whose higher trypomastigotes, trypomastigotes without corresponding increase levels.
