摘要: Abstract Rhesus monkeys are relevant models for human diseases and transplantation. In each case, a complete understanding of these requires knowledge macaque MHC. Due to high polymorphism multiple genes per haplotype, it has been difficult develop rapid typing method rhesus monkey MHC class I. We developed simple PCR-SSP strategy Mamu-A locus typing. Fifty-two were included in the study. Six allel-specific primer pairs designed based on published gene sequences. Allele-specific PCR products ranged size from 346 788 bp; 5′ 3′ allele specific primers located second third exons, respectively. Specific product gel purification was followed by direct sequencing, without subcloning, both directions. Our data showed variability number alleles ranging 1 4 genotype. The highest frequencies observed Mamu-A∗02 , -A∗04 -A∗03 alleles. Thus, we report here first -03 -04 -05 -06 -07 array I This technique appears be highly reproducible discriminatory detecting this subset A monkeys.
