摘要: Ion microscopy was employed to investigate intracellular total calcium concentrations and influx, efflux in resting antigen-stimulated tumor mast cells (RBL-2H3 cells). The nucleus, a perinuclear region which included the Golgi apparatus (Golgi region), remaining cytoplasm were spatially resolved with Cameca IMS-3f ion microscope cryogenically prepared cells. In nucleus contained about 0.60 mM, 1.2 1.0 mM calcium. Antigen stimulation of rat basophilic leukemia resulted significant loading all three cellular compartments. absence extracellular loss from Influx measured simultaneously stimulated by using stable 44Ca solution, imaging mass 40 determine native (40Ca) 44 localize that entered cell solution. After 10-min incubation, 0.240 fmol per had been replaced 44Ca, amounts 33% If antigen present during this incubation there an additional 0.229 40Ca added gain 0.476 cell, corresponds net increase 0.247 fmol.
