摘要: A fluorescent 16S rRNA oligonucleotide probe was developed specific to the family Enterobacteriaceae. The target sequence is 5'-CUCGCGAGAGCAAGCGG-3', corresponding positions 1273 1289 on Escherichia coli rRNA. Specificity of Enterobacteriaceae confirmed using representative members 20 genera comprising and other selected bacteria. Successful application an environmental habitat demonstrated wastewater samples collected from primary secondary effluent a treatment plant (WWTP). DAPI EUB338, domain Bacteria, were used enumerate total prokaryotic cells, respectively, in effluent. probing efficiency, defined as number bacteria enumerated with EUB338 divided by organisms DAPI, both approximately 94%. relatively high efficiency indicates copy sufficiently within ensure detectable image. can potentially be engineering profession alternative standard coliform test tool evaluate effect upstream processes at WWTP disinfection performance.
