摘要: Reovirus selectively replicates in and destroys cancer cells with an activated Ras signaling pathway. In this study, we evaluated the feasibility of using reovirus (serotype 3, strain Dearing) as antihuman colon ovarian agent. vitro studies, infection human cell lines was assessed by cytopathic effect detected light microscopy, [(35)S]Methionine labeling infected for viral protein synthesis progeny virus production plaque assay. We observed that efficiently all five (Caco-2, DLD-1, HCT-116, HT-29, SW48) four (MDAH2774, PA-1, SKOV3, SW626) which were tested, but not a normal line (CCD-18Co) or (NOV-31). also activity elevated compared lines. animal models, intraneoplastic well i.v. inoculation resulted significant regression established s.c. tumors implanted at hind flank. Histological studies revealed vivo restricted to tumor cells, whereas surrounding tissue remained uninfected. Additionally, i.p. xenograft model, inhibition ascites formation survival animals treated live significantly greater than control mice UV-inactivated reovirus. ex primary surgical samples confirmed, further demonstrating potential therapy. These results suggest holds promise novel agent
