Use of a panel of congenic strains to evaluate differentially expressed genes as candidate genes for blood pressure quantitative trait loci.

摘要: Candidate gene(s) for multiple blood pressure (BP) quantitative trait loci (QTL) were sought by analysis of differential gene expression patterns in the kidneys a panel eight congenic strains, each which carries different low-BP QTL allele with genetic composition that is otherwise similar to hypertensive Dahl salt-sensitive (S) rat strain. First, genes differentially expressed one-month-old S and salt-resistant (R) rats identified. Then, Northern filter hybridization was used examine these strains. Finally, their chromosomal location determined radiation hybrid (RH) mapping. Seven out 37 mapped regions carrying BP QTLs, but only one genes, L-2 hydroxy acid oxidase (Hao2), showed strain-specific pattern kidney predicted its location. This data suggests Hao2 should be examined as candidate chromosome 2 (RNO2) QTL.
