摘要: Brevican is a nervous system-specific chondroitin sulfate proteoglycan that belongs to the aggrecan family and one of most abundant proteoglycans in adult brain. To gain insights into role brevican brain development, we investigated its spatiotemporal expression, cell surface binding, effects on neurite outgrowth, using rat cerebellar cortex as model system. Immunoreactivity occurs predominantly protoplasmic islet internal granular layer after third postnatal week. Immunoelectron microscopy revealed localized close association with astrocytes form neuroglial sheaths glomeruli where incoming mossy fibers interact dendrites axons from resident neurons. In situ hybridization showed synthesized by these themselves. primary cultures astrocytes, detected cells. Binding assays exogenously added several immortalized neural lines have binding sites for core protein. These recognize C-terminal portion protein are independent hyaluronan. results indicate retained their an interaction involving Purified inhibits outgrowth granule neurons vitro, activity requires chains. We suggest presented may be controlling infiltration maturing glomeruli.
