摘要: Cultures of primary tumors are very useful as a personalized screening system for effective therapeutic options. We here describe an method reproducing human colon through culture and mouse xenograft model. A total 199 tumor cultures were successfully established under optimized conditions to enrich cells expand it long-term storage in liquid nitrogen. To examine whether these stored retained original properties, fifty xenografted into NOD-SCID mouse. Histological marker analysis four representative xenografts revealed that all the its characteristics. Oncomap further showed no change major mutations xenografts, confirming our faithfully reproduced tumors. drug sensitivity assay two hypersensitive oxaliplatin rather than 5-FU, which was used patients, suggesting option. thus present effective, reproducible preclinical model testing various options cancer patients.
