摘要: The precise mechanism responsible for the frequent overexpression of cyclinD1 in human head and neck squamous cell carcinoma (HNSCC) is not known. In view fact that signal transducers activators transcription 3 (Stat3) often activated HNSCC cells, we examined effects Stat3 on cyclin D1 expression proliferation YCU-H891 line displays constitutive activation Stat3. Expression a dominant negative construct cells inhibited proliferation, promoter activity, cellular levels mRNA protein. antiapoptotic Bcl-2 Bcl-X(L) proteins were also inhibited. 51 primary tumor samples from patients with p.o. tongue, there was significant correlation between increased form Stat3, phosphorylated-Stat3, (P < 0.0001). Increased phosphorylated-Stat3 associated lower survival rates 0.01). This study provides first evidence HNSCC, plays causative role D1, clinical studies, may provide novel prognostic factor.
