摘要: An on-line solid-phase extraction (SPE)-HPLC method was developed for simultaneous screening of nicardipine and amlodipine in human plasma. A short monolithic poly(glycidyl methacrylate-co-ethylene glycol dimethacrylate) [p(GMA-EDMA)]-based weak cation-exchange (WCX) column prepared employed as the selective sorbent, which exhibited good permeability biocompatibility. During SPE protocol, high-abundance proteins (human serum albumin, immunoglobulin G, transferrin) most matrixes plasma were fast removed while effectively trapped on this column. Furthermore, WCX sorbent could be continuously reused more than 300 times without obvious changes analytes cleanup. The proposed linear over a range 0.5-50.0 ng mL(-1) both with regression coefficient greater 0.998, limit detection (LOD) each analyte 0.2 mL(-1). Validation assays also demonstrated acceptable precision adequate recovery quantitative Real samples from hypertensive patients receiving dosing 5mg antagonists examined by using method. Results indicated that SPE-HPLC applied simultaneously monitoring clinical samples.
