摘要: The heme peroxidase HPX15 is an evolutionary conserved anopheline lineage-specific gene. Previously we found that this gene present in the genome of nineteen worldwide distributed different species Anopheles mosquito and its orthologs are absent other mosquitoes, insects or human. In addition, 65-99% amino acid identity among these 19 permitted us to hypothesize functional aspects might be also anophelines. study, stephensi AsHPX15 mainly expressed midgut highly induced after uninfected Plasmodium berghei infected blood feeding. RNA interference (RNAi)-mediated silencing drastically reduced number developing P. oocysts. An antiplasmodial nitric oxide synthase (NOS) was thirteen fold silenced midguts when compared unsilenced controls. Interestingly, induction immunity absolute agreement with A. gambiae. gambiae, AgHPX15 catalyzes formation a dityrosine network at luminal side suppresses activation against bolus bacteria. Thus, low zone created by mechanism indirectly supports development inside lumen. These indistinguishable behaviors homology indicates potent target manipulate it will open new frontiers field malaria control.
