Dietary administration of Nexrutine inhibits rat liver tumorigenesis and induces apoptotic cell death in human hepatocellular carcinoma cells

摘要: Epidemiological studies suggested that plant-based dietary supplements can reduce the risk of liver cancer. Nexrutine (NX), an herbal extract from Phellodendronamurense, has been shown to have anti-inflammatory, anti-microbial and anti-tumor activities. In present study, we potential NX against Solt-Farber model with elimination PH, rat tumor induced by diethylnitrosoamine (DEN) as carcinogen 2-acetylaminofluorene (2-AAF) co-carcinogen. The elucidation mechanistic pathways was explored in human cancer cells. Dietary intake significantly decreased cell proliferation inflammation, well increased apoptosis sections DEN/2-AAF-treated rats. Moreover, (2.5–10 μg/ml) exposure viability cells modulated levels Bax Bcl-2 proteins levels. treatment resulted cytochrome-c release cleavage caspases 3 9. addition, expression CDK2, CDK4 associated cyclins E1 D1, while up-regulated p21, p27 p53 expression. also enhanced phosphorylation mitogen-activated protein kinases (MAPKs) ERK1/2, p38 JNK1/2.
