摘要: Enhancers are cis-act ing elements that activate transcription in higher eukaryotes independently of their position or orientation relative to the promoter they activate1. The mechanisms by which enhancers poorly understood, part because, with exception glucocorticoid receptor2, proteins directly interact have not been purified, nor genes encoding them cloned3–5. upstream regulatory region (URR) immediately precedes early bovine papillomavirus type 1 genome (BPV) has enhancer activity when it is activated a trans-acting gene product BPV E2 open reading frame (ORF) (Fig. 1)6. It known whether this enhancement represents direct indirect effect on URR. We used an peptide expressed bacteria and DNA-protein complex immunoprecipitation assay study E2-mediated show here binds four specific sites URR, one site human (H)PV16 All binding contain related sequence nucleotides; 23 base pair (bp) fragment containing can specifically prevent protein E2–URR interaction may therefore represent useful model system for studying mechanism transcriptional enhancement, as both effector its target be purified genetically manipulated.
