摘要: PURPOSE. The immunogenic characterization of amniotic membrane is still unknown. This study was designed to examine the immunogenicity human membrane, by using experimental xenotransplantation models. METHODS. Anti-human class I, II, and Fas ligand monoclonal antibodies were used against cryopreserved cell viability tested for membrane. Amniotic membranes then transplanted limbal area, intracorneal space, under kidney capsule. scores transparency neovascularization after transplantation recorded slit lamp microscopy. Host infiltration examined hematoxylin-eosin or immunohistochemical staining. Control grafts skin grafts. RESULTS. Strong I expression observed in epithelium, mesenchymal cells, fibroblasts Some fibroblast cells unexpectedly expressed II antigen. ligand‐positive also detected stroma. Approximately 50% epithelial several months viable. In transplantation, although some CD4 1 CD8 T surrounded graft, response mild. all grafted accepted clear, without host infiltration. contrast, rejected within 3 weeks transplantation. capsule, extremely few vessels infiltrated membrane; however, more tissues CONCLUSIONS. seems be immune-privileged tissue contain immunoregulatory factors, including HLA-G ligand. may useful supplement corneal collagen, it applied not only ocular surface but intracorneally.
