摘要: BACKGROUND Heparin is known to have antithrombotic, anticoagulant, and antiproliferative effects. We hypothesized that chronic subcutaneous and/or direct intramural administration of heparin would reduce restenosis inhibit plaque growth after balloon angioplasty. METHODS AND RESULTS Focal atherosclerosis was induced bilaterally in the femoral arteries 59 rabbits by air desiccation intimal injury a 2% cholesterol diet. After angioplasty, were assigned one four treatment groups. Control (n = 21) received no additional heparin. A second group 20 treated with porous delivered (1,500 units) directly into arterial wall. third 29) (350 units.kg-1.day-1) for 28 days, fourth 23) Quantitative angiography showed modest reduction (defined as change minimal luminal diameter from immediately angioplasty days) compared control (0.32 +/- 0.18 versus 0.58 0.34 mm, p < 0.01); however, not improved at days before angioplasty. Intramural delivery catheter confirmed use fluoresceinated animal. Angiographic reduced controls (0.61 0.54 NS). Blinded planimetric analysis histological sections differences cross-sectional area narrowing atherosclerotic plaque, area, or plaque/media ratio among groups. CONCLUSIONS Chronic results angiographic this model; absolute percent different can be an balloon, but does model.
