FGF1 and IGF1-conditioned 3D culture system promoted the amplification and cancer stemness of lung cancer cells

摘要: Abstract Lung cancer stem cells (LCSCs) are considered as the cellular origins of metastasis and relapse lung cancer. However, routine two-dimensional culture system (2D-culture) hardly mimics growth functions LCSCs in vivo therefore significantly decreases stemness activity LCSCs. In this study, we constructed a special BME-based three-dimensional (3D-culture) to amplify in human adenocarcinoma cell line A549 cells found 3D-culture promoted enrichment amplification displaying higher proliferation potential invasion activity, but lower apoptosis. The expression secretion levels FGF1 IGF1 were dramatically elevated compared 2D-culture. After growing IGF1-conditioned 3D-culture, proportion with specific phenotypes increased that conventional 3D suspension system. Further results indicated dependent on MAPK signaling pathway. Our data firstly established factors-conditioned for demonstrated effects promoting which might provide feasible model in vitro both mechanism study translational research
