摘要: SUMMARY Glioblastoma multiforme (GBM) is the most common primary malignant brain tumor in adults and carries poorest prognosis despite aggressive multimodal therapy. The majority of GBMs develop de novo (primary) with a short clinical history, while secondary through progression from preexisting lowergrade precursor gliomas show distinct genetic expression profiles including high frequency isocitrate dehydrogenase 1 (IDH1) mutations, already present lesions. Large-scale integrative genomic studies provided new view that are remarkable molecularly heterogeneous tumors identified molecular entities may lead to different therapeutic approaches. Although being restricted intracranial compartment, associated global immunosuppression. Better understanding immune response growing immunologically microenvironment mechanisms by which they escape even suppress it will accelerate development more effective immunotherapies. This review summarizes current knowledge regarding alterations signaling pathways critical biology GBMs, few developing local systemic GBM-induced immunosuppression, role GBM stem cells.
