摘要: Background: Colorectal adenomatous and, probably, hyperplastic polyp development requires epithelial remodelling and stratification, with loss of E-cadherin expression implicated in adenoma formation. We have shown that P-cadherin, normally expressed stratified epithelia placenta, is aberrantly disturbed architecture associated colitis. Aims: (i) To investigate the role P-cadherin colonic (ii) ascertain whether independent or correlated its proteins— E-cadherin, β-catenin, γ-catenin. (iii) determine if functional regarding catenin binding polyps. Methods: Expression localisation cadherins (E- P-) their catenins (β- γ-) were determined aberrant crypt foci (ACF), polyps morphology (hyperplastic serrated adenomas), by immunohistochemistry, western blotting, mRNA situ hybridisation. Assessment cadherin-catenin was evaluated co-immunoprecipitation. Adenomatous polyposis coli (APC) mutation assessed polyps. Results: from ACF through to polyps. Alterations occurred later, variant patterns ACF, adenomas, present adenomas regard binding, APC mutational status. Conclusions: earliest morphologically identifiable stage colonocyte transformation, prior changes catenin, expression/mutation. alone does not predict tissue morphology, such catenins.
