摘要: Complement receptor lymphocytes (CRL) were isolated from lymph nodes of rabbits immunized 6 days earlier with keyhole limpet hemocyanin. Isolation was accomplished by rosetting sheep erythrocytes sensitized antibody and horse complement (EAC) followed Centrifugation on Ficoll-Hypaque (F-H). The CRL population comprised about 30% the starting LNC highly enriched for surface immunoglobulin (SIg) bearing cells. By criterion [3H]thymidine incorporation, responded poorly to Concanavalin A (ConA), but extensively goat anti-rabbit Fab′ (AFab′) ; indeed, better than unfractionated AFab′. Upon addition KLH, produced much less total Ig LNC. KLH-challenged T cells prepared anti-Fab′ columns also synthesized minimal amounts Ig. Antigenic challenge a mixture 3 5 times more sum individual CRL- cell-mediated syntheses.
